Interpretation of mass spectrometry (MS) data can help solve problems. The goal of this 2.5 day course is to improve your mass spectral interpretation skills. This course is followed by a three day "Advanced Mass Spectral Interpretation" course, PACS course number 20. Your MS success needs effective data interpretation skills. This popular course is designed for providers and users of mass spectral data. It is designed to improve the productivity and quality of results from your MS lab and/or to facilitate wise decisions for your firms use of MS data. If you have a present or future need to use MS to help solve problems but require basic principles and some directions, this course is for you. Plan to bring your calculator or laptop computer. This course is a "hands on" problems working course. A course outline is below.

The course leader is Dr. Henry Nowicki; he has well over thirty years of MS experiences in many problem solving application areas: environmental sciences, clinical, toxicology, forensics, legal and illegal drugs, natural products, metabolism, food sciences, R&D, biotechnology, and others. A required text is "Interpretation Of Mass Spectra", fourth edition by Fred McLafferty and F. Turecek. This book must be brought to your PACS course and can be purchased from: University Science Books, 20 Edgehill Road, Mill Valley, CA 94941 or phone (415) 332-5390 and FAX (415) 332-5393. This course will cover most of the problems in the first five chapters and additional selected subjects.

Data acquisition strategy – SIM and full scan
MS components – sample introduction, ion source, separator, detector and computer
Electron impact (EI) and chemical ionization (CI) and when to use each
Tuning and resolution/sensitivity trade off
Full scan and selected ion monitoring and when to use each
Major atom types for organic mass spectrometry
Isotopes and mass spec use – natural abundances and man made molecules
Importance of molecular ion
Molecular ion tests and intensity relationship to parent ion structure
Hierarchy and predictability of electron loss location from parent ion
Basics of spectral interpretation and homework problems
DAY 2
Development of MS interpretation skills to match needs
Solve and discuss day 1 MS homework interpretation problems
Simple cleavage and relative ion stabilities
Odd-even(low mass ion series-molecular ion relationship) rule
Nitrogen rule odd mass molecular ion
Importance of neutral loss for MS interpretation
Characteristic ion patterns and diagnostic ions
McLafferty hydrogen rearrangement exercises
Carbon number calculation methods and their uses
Solve mass spectral interpretation problems and discussion
Use F.W. McLafferty’s book to solve problems, homework problems
DAY 3
Applications for mass spectrometry
Isotopic cluster recognition practice
Polycyclic aromatic hydrocarbons
Saturated and unsaturated aliphatic hydrocarbons
Alcohols and sulfur isosteric replacement
Aldehydes, carboxylic acids, and esters
Ketones and ethers
Aromatic organics
Halogenated organics
Hydrogen rearrangements (McLafferty’s and plus one)
Solve mass spectral interpretation problems and discussion
